In 1980, a study of geographic distribution patterns and geographic affinities of benthic algae of temperate Pacific South America (5-55 ºS) provided a first characterization of these coasts. High endemism and limited floristic exchange with the Tropical Pacific and with islands in the South Pacific at various distances from the continent were two outstanding features of the marine flora. A partial blockage of species exchange partially accounted for a peculiar latitudinal pattern of species richness that increases with increasing latitudes. Isolation and survival of Yersinia enterocolitica in ice cream at different pH values, stored at -18°c BJM

The presence of Yersinia enterocolitica was investigated in 203 samples of industrial (123) and non-industrial ice cream (80). Two Y. enterocolitica strains were isolated from non-industrial ice cream, which suggests the possibility of post-manufacturing contamination. One strain was typed as B:1A, O: 3,50,51; lis Xz, while the other one was biotyped as: B:1A but not serologically typed. Characterization of a highly thermostable alkaline phosphatase from the euryarchaeon Pyrococcus abyssi ArchiMer

This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Isolation, properties and kinetics of growth of a thermophilic Bacillus BJM

A strictly aerobic, thermophilic, spore-forming bacteria was isolated from a soil sample collected in Campos dos Goytacazes City, Rio de Janeiro, Brazil. The cells of this organism were Gram-positive, catalase positive, actively motile, 1.2 µm wide and 5.3 µm long. Growth occurred at pH values ranging from 6.5 to 9.0, and optimum growth occurred at about pH 7.0. The optimum temperature for growth was around 55ºC, and the upper temperature limit for growth was around 70ºC. Yeast extract stimulated growth on glucose and was obligately required at supraoptimal temperatures (65ºC). This work was conducted on rats in two premises located in Niterói and Rio de Janeiro, Brazil. One is classified as conventional controlled and the other, conventional. The objective of the present study was to detect the presence of Mycoplasma pulmonis in animals with symptoms of respiratory disease and low reproductive performance. In the conventional controlled premises, 16 rats of Wistar-Furth strain were necropsied while in the conventional premises necropsy was performed on 12 rats of Hooded Lister strain. The clinical samples of lungs, trachea, oropharynx, middle ear, uterus and ovaries were subjected to culturing while the sera were tested for antibody detection. Two banana supply chains (BSC), one in Indonesia (BSC1) and one in Australia (BSC2), are described based on case study research conducted in 2002. BSC1 sources bananas from small farmers for distribution through traditional markets in Jakarta. BSC2 includes farmers supplying to both major and independent retailers in Sydney through a wholesale agency with some farmer ownership. Farmers in both chains live in relative isolation from the main markets for their bananas. Prices for their bananas depend on the market prices in the city, upon which they have little control. The aim of this study was to adapt a mechanical procedure for the isolation of intact preantral follicles from Cebus apella ovaries. The interval effect of serial sections of the tissue chopper was tested on a number of preantral follicles isolated from ovaries (n=6) of three C. apella females, two prepubertal and one adult. Ovaries were divided into four equal parts and fragmented with a tissue chopper, adjusted for serial sections at intervals of 250, 500, 750 and 1,000µm, respectively. Brucella spp. isolation from dogs from commercial breeding kennels in São Paulo state, Brazil BJM

Dogs from 12 commercial breeding kennels were submitted to clinical investigation and laboratorial tests for diagnosis of Brucella spp. infection. The sampling was carried out between April 2000 and February 2002 and the laboratorial tests employed were agar gel immunediffusion test (AGID) and blood culture. From 171 dogs examinated, 39 (22.8%) showed at least one clinical sign compatible with brucellosis, 58 (33.91%) were AGID positive and 24 (14.03%) were positive by blood culture. Isolation of bovine coronavirus (BCoV) in monolayers of HmLu-1 cells BJM

Isolation of BCoV was performed in monolayers of HmLu-1 cells, using 20 fecal samples from clinical cases of diarrhea in calves. Samples were positive for BCoV by means of hemagglutination / hemagglutination inhibition (HA/HI). Up to the fifth serial passage, 13 of these isolates presented syncytial cytopathic effect, similar to Kakegawa standard strain. When isolates were submitted to seroneutralization with gammaglobulin anti-bovine coronavirus, 8 of them were considered to be positive, once cytopathic effect was neutralized. The occurrence of Salmonella in a samples of 40 imported day-old duckling flocks was assessed from 1998 to 2003 according to the guidelines of the Brazilian National Poultry Health Program (Programa Nacional de Sanidade Avícola-PNSA). The pathogen was recovered from 26 flocks (65.0%). The most common serovars were S. Saintpaul and S. Kottbus. Up to four serovars were isolated from a single flock. Transportation box swabs (82.6%) and yolk sac pool (47.1%) showed the highest and the lowest frequency of Salmonella isolation, respectively. The high percentage of Salmonella isolation from imported day-old ducklings causes concern because of the zoonotic potential of this agent and its economical importance to commercial poultry breeding.

Reservoir competence for the Newcastle Disease virus (NDV) was evaluated in sparrows (Passer domesticus, Linnaeus 1758) captured on a commercial poultry farm and a chicken hatchery in the State of Pernambuco, Northeastern Brazil. A total number of 103 birds collected from a poultry farm (24/103) and a chicken hatchery (79/103) were examined. Hemagglutination inhibition tests, isolation, and viral characterization were performed in all samples collected from each bird. Titers ranging from 1:2 to 1:64 were detectable in 10.68% of sparrows, but positive serology and viral isolation were obtained only from sparrows captured at the hatchery. A phospholipase A2 has been isolated from Bothrops jararacussu venom from snakes that inhabit the northeast region of Argentina. The present study describes in vivo and in vitro biological activities of phospholipase A2 from B. jararacussu as well as isolation details of its. Venom was obtained by milking of adult snakes which were housing in wood reptile cages of varying dimensions in heated (20-30ºC) rooms. El objetivo del presente estudio fue obtener un cultivo puro de la bacteria ruminal Streptococcus bovis a partir de líquido ruminal de ovinos alimentados con una dieta alta en sorgo (70 % en base seca). Además, se analizó su actividad fermentativa (producción de AGV y lactato) y el proceso de liofilizado como método de conservación, y se realizaron ensayos para expresar su bacteriófago. Para el aislamiento se usó un medio comercial para Streptococcus spp. (Merck) preparado bajo anaerobiosis.
